چکیده

Enzymes that are capable of hydrolyzing ribonucleoside 2’, 3’cyclic phosphates but that do not attack intemucleotide bonds are available from various sources (1). Several of these enzymes convert ribonucleoside 2’,3’-cyclic phosphates to the corresponding ribonucleoside 2’-phosphates. The first die&erase of this type was studied in calf spleen by Whitfeld, Heppel, and Markham (2). Davis and Allen (3) later described a highly specific enzyme from beef pancreas that converted purine and pyrimidine 2’,3’-cyclic phosphates to the 2’-phosphates. In a previous report from this laboratory (4), it was briefiynoted that rabbit brain extracts catalyzed a rapid hydrolysis of these compound8 and that the product of hydrolysis of adenosine 2’,3’-cyclic phosphate was adenosine 2’-phosphate. This paper describe8 further studies concerning the properties and the specificity of this enzyme in the central nervous system.

Enzymatic hydrolysis of adenosine 3',5'-phosphoric acid.

During recent years the work of Sutherland and his associates (l-3) on the liver phosphorylase system has done much to clarify the mechanism of epinephrine-induced hyperglycemia. These workers have shown that epinephrine stimulates glycogenolysis by increasing the level of active phosphorylase in dog liver slices (3, 4) and in cell-free homogenates (5). The effect of epinephrine was shown to be...

Non-canonical 3′-5′ Extension of RNA with Prebiotically Plausible Ribonucleoside 2′,3′-Cyclic Phosphates

Ribonucleoside 2',3'-cyclic phosphates (N>p's) are generated by multiple prebiotically plausible processes and are credible building blocks for the assembly of early RNA oligomers. While N>p's can be polymerized into short RNAs by non-enzymatic processes with variable efficiency and regioselectivity, no enzymatic route for RNA synthesis had been described. Here we report such a non-canonical 3'...

Identification of 2':3'-cyclic nucleotide 3'-phosphodiesterase in the vertebrate retina.

The enzyme, 2':3'-cyclic nucleotide 3'-phosphodiesterase (2':3'-cNMP-3'-ase) has been used as a marker in the nervous system for the presence of myelin membrane or myelin-producing glial cells. In this study, goldfish and bovine neural retinas are found to have high levels of such a diesterase activity. Analysis of retinal tissue incubated with 2':3'-cAMP shows only 2'-AMP as the reaction produ...

A New Cyclic Phosphodiesterase Having a 3'-nucleotidase Activity from Escherichia Coli B. Ii. Further Studies on Substrate Specificity and Mode of Action of the Enzyme.

A number of phosphodiesterases which can hydrolyze ribonucleoside 2’, 3’-cyclic phosphates have been demonstrated in several biological materials (l-6). The preceding paper (7) presented evidence which strongly suggests the existence of a novel enzyme, designated temporarily as a cyclic phosphodiesterase, in the 78,000 x g supernatant of Escherichia coli B. At the same time a 3’-nucleotidase (E...

Activation of cardiac phosphorylase b kinase.

Nonactivated, partially purified bovine heart phosphorylase b kinase was activated by incubation with adenosine triphosphate and Mg+f ion prior to assay. Adenosine 3’ ,5’-cyclic pgosphate increased the rate of activation, but there was no absolute requirement for the cyclic nucleotide. Activation was particularly prominent when the subsequent assay for kinase activity was carried out at pH 7.0,...
